青海省部分地区牛的皮蝇蛆分子分类学研究
摘要
本研究旨在分析感染牛的皮蝇蛆分子分类地位.通过聚合酶链式反应(PCR)对4株感染牛的皮蝇蛆的线粒体CO1基因片段进行扩增,克隆和测序 ,并将其序列与GenBank中的中华皮蝇H.sinense(AY350769)和纹皮蝇H.lineatum(AF497762)的基因序列进行了比对分析,构建遗传进化树.结果显示,扩增出的CO1基因片段为688bp,利用ClustalX进行多序列比对时发现都有不同程度的差异,差异1个碱基至几个碱基不等,同源性从96.8%到99.7%.其中华皮蝇与玉树珍秦乡(YS)、唐古拉镇(TGL)聚集交错在一起并与海晏青海湖乡聚为一大类,说明他们的亲缘关系比较近;纹皮蝇H.lineatum(AF497762)与青海民和满坪乡皮蝇聚一支;这与青海不地区牛的皮蝇蛆形态鉴定和地理分布一致.系统发育树与这些品种的育成史、分化及地理分布相符.

This study aims to analysis of bovine leather bots molecular classification status.By polymerase chain reaction (PCR) of four strains of bovine leather bots mitochondrial CO1 gene amplification,cloning and sequencing,and the skin in the sequence from GenBank flies H.s inense (AY350769) and grain leather fly H.l ineatum (AF497762) gene sequence has carried on the comparative analysis,construction of genetic evolutionary tree.Results show that the amplification of CO1 gene fragment of 688 bp,multiple sequence alignment using the ClustalX and found all have different degree of difference,difference of one base to a few bases,homology from 96.8% to 99.7%.Skin of China flies and yushu Jane qin township (YS),tangula (TGL) gathered staggered together with sea taking qinghai reaches for a broad categories,their close relatives;Grain leather fly H.l ineatum (AF497762) and qinghai minhe ping township leather fly together a; The cow leather and qinghai area bots morphological identification and distribution agreement.Phylogenetic tree with these varieties breeding history,differentiation and geographical distribution.
