摘要:本实验以新鲜鳙鱼的肌肉、肝脏、尾鳍为实验材料,采用75%乙醇保存;利用裂解液和蛋白酶K消化新鲜鳙鱼肌肉、肝脏、尾鳍,通过苯酚-氯仿提取法提取基因组DNA,再进行琼脂糖凝胶电泳和分光光度法检测.结果表明,苯酚法得到的DNA浓度高,质量好,且相对成本低.综合来说:鳙鱼肌肉、肝脏、尾鳍DNA提取的结果是:肝脏>肌肉>尾鳍.新鲜组织比75%乙醇保存的组织提取的DNA效果好.稀释20倍后,鳙鱼分光光度法测得的浓度在6.9.8μg•mL – 1左右,A260/A280在1.712-1.983.

Abstract:this experiment need fresh Aristichthys keeped in the 75% ethyl alcohol ; digesting the fresh Aristichthys nobilis'muscle ,liver and fin by lysate and Proteinase K,extracting genome DNA by phenic acid ,then proceed the inspection of agarose gel electrophoresis and spectrophotography.result showing:phenic method can get more condensed ,higher quality and low-cost DNA.all in all,the result of inspection for nobilis'muscle ,liver and fin is:live>muscle>fin.DNA distilled from fresh tissue is better than that preserved in ethyl alcohol.diluting for 20 times,condensity of spectrophotography is about 6.9.8μg•mL – 1,A260/A280 is 1.712-1.983.
