摘 要:本文研究了合成16C14-18系列酰胺类分子,以L-丝氨酸高碳烷基酯盐酸盐和棕榈酸为原料,以吡啶为催化剂,通过酰胺化反应达到合成N-十六酰基-L-丝氨酸高碳烷基酯(神经酰胺拟合物)为目标产物.由酯化反应的单因素实验,探讨了反应时间,反应温度,原料配比对酰胺化反应产率的影响.由正交实验确定了最优工艺条件为:n(L-丝氨酸高碳烷基酯盐酸盐):n(棕榈酰氯):n(吡啶)=1:1.3:3.5,反应温度为60oC,反应时间为3.5h.在此条件下,转化率可达到89%.通过采用红外光谱表征,用ELSD-HPLC测得含量,用紫外光谱和熔点测定来检测物理性质.通过进一步的放大实验,验证了上述工艺条件是稳定可靠的,且操作工艺简便,为以后的实际应用提供了指导意义.

Summary:The paper studies the synthesis of 16C14-18 series lactam molecules,With L-serine-carbon alkyl ester hydrochloride and palmitic acid as raw materials.Achieved by amidation Synthesis of N-palmitoyl-L-serine-carbon alkyl ester (ceramide fitting material) as the target product.The effects of reaction time,reaction temperature,the ratio of raw materials for the amidation reaction yield were Investigate by the esterification reaction of single factor experiments,Determined by the orthogonal experiment the optimal process conditions:n (L-serine-carbon alkyl ester hydrochloride):n (palmitoyl chloride):n(pyridine) = 1:1.3:3.5,the reaction temperature was 60oC,and the reaction time was 3.5h.In this condition,the conversion rate reached 89%.The product was characterized by using Infrared spectroscopy,the product concent was determined by ELSD-HPLC,and the physical properties were detected by UV and melting point measurement.Through further amplification experiments,the process conditions was proved to be stable and reliable,and the operation process is simple,it will provide guiding significance for practical application.
