本实验利用扩增性rDNA限制性酶切片段分析(ARDRA) 技术,通过建立16s rDNA克隆文库对大庆油田油层微生物群落进行初步分析.扩增性rDNA限制性酶切片段分析(ARDRA) 技术是依据原核生物rDNA序列的保守性,将扩增的rDNA片段进行酶切,通过酶切图谱来分析菌间多样性.分析结果表明:大庆油田采油二场原油微生物群落以Acinetobacter(不动杆菌属)为主,还有一定量Pseudomonas(假单胞菌属)、Hyphomonas(生丝单胞菌属)、Oceanibaculum、Maricaulis属菌.由四个时间点菌体DNA建库分析后相互比较可看出,随着培养时间的延长和转接传代的延续,Acinetobacter在群落中所占比例减少,其他种属的细菌逐渐增多.

In this study,the use of amplified rDNA restriction fragment analysis (ARDRA) technology,through the establishment of 16s rDNA clone library of the Daqing oil field reservoir preliminary analysis of microbial communities.Amplified rDNA restriction fragment analysis (ARDRA) technique is based on the prokaryotic rDNA sequence of the conservative nature of the amplified rDNA fragments were digested by restriction mapping to analyze the diversity among bacteria.The results show that:Daqing oilfield microbial communities in two oil fields in order to Acinetobacter (Acinetobacter) as the main,there is a certain amount of Pseudomonas (Pseudomonas),Hyphomonas (raw silk Aeromonas spp),Oceanibaculum,Maricaulis is a bacteria.From the four time points analysis of bacterial DNA Database comparisons can be seen,with the incubation time of the extension and continuation of transit passage,Acinetobacter in the community to reduce the proportion of other species of bacteria gradually increased.
