这说明你的操作没有问题,只是你的样本确实是阴性的.至于你的样本为什么是阴性,要看你的阳性对照到底是哪里改变了.

If you included positive and negative controls, and if your positive is positive and negative is negative, then whatever the results you obtained is true. The possible reason why your samples become negative is as followed:
1. you have DNase contamination with your samples, degradating your samples DNA.
2. You did not included enough template DNA in your second test.
3. check whether you included right primers and whether you included enough.
